The neuropathological basis of the disease involves production of pathogenic amyloid-β (Aβ) oligomers from amyloid precursor protein (APP), hyperphosphorylated tau, and synapse loss resulting in a “dying back” neuropathy and ultimately neuron death in both cortical and sub-cortical regions (Katzman, 1986; Arnold et al., 1991).
